Tofacitinib is a Janus kinase (JAK) inhibitor used in the treatment of rheumatoid arthritis (RA). It is typically prescribed for moderate to severe cases of RA when other disease-modifying antirheumatic drugs (DMARDs) have not provided adequate relief.

Mechanism of Action: Tofacitinib works by inhibiting specific enzymes known as Janus kinases. These enzymes play a role in the signaling pathways that lead to inflammation. By inhibiting these enzymes, tofacitinib helps reduce inflammation and slow down the progression of RA.

Dosage: The usual recommended dosage of tofacitinib for rheumatoid arthritis is 5 mg taken orally twice daily. In addition to the standard formulation, there is also an Tofacitinib 11 mg tablet extended-release version available  that is taken once daily.

Side Effects: Tofacitinib, like any medication, can have side effects. Common side effects may include upper respiratory tract infections, headache, and high blood pressure. Serious side effects may include an increased risk of serious infections, certain cancers, and liver problems.
